Overview

MDNA11 is a next-generation, long-acting recombinant interleukin-2 (IL-2) immunotherapeutic engineered as a "Superkine" to preferentially bind the IL-2 beta receptor (IL-2Rβ/CD122) on immune cells. It is designed to activate and expand anti-tumor immune effector cells—specifically CD8+ T-cells and natural killer (NK) cells—while minimizing stimulation of regulatory T-cells (Tregs), which can suppress anti-tumor immunity. MDNA11 achieves this selectivity by incorporating seven specific mutations that abrogate binding to the IL-2 alpha receptor (CD25), fused genetically to human albumin for improved pharmacokinetics and tumor targeting. The drug is administered intravenously and is being developed primarily for advanced solid tumors, both as monotherapy and in combination with checkpoint inhibitors such as pembrolizumab.
