Overview

MDNA132 is a modified interleukin-13 (IL-13) Superkine engineered to bind with extremely high affinity—up to 16 million times greater than native IL-13—to the “dummy” IL-13 receptor subunit alpha-2 (IL-13Rα2). This receptor is overexpressed in many solid tumors but minimally expressed in normal tissues. By targeting IL-13Rα2, which acts as a decoy receptor and is associated with aggressive tumor phenotypes and poor survival outcomes, MDNA132 enables highly selective accumulation within the tumor microenvironment. The drug’s design allows for precision immunotherapy approaches that can selectively deliver therapeutic payloads or engage immune cells directly at the site of tumors that are resistant to conventional immunotherapies. Medicenna has also developed fusion proteins based on this platform (e.g., Anti-mCD3-MDNA132), combining the superkine with an anti-CD3 antibody to recruit immune cells for direct tumor cell killing[1][3][5].
