Overview

MDNA213 is a novel engineered protein classified as an "IL-13 Superkine," designed to selectively and with high affinity bind the interleukin-13 decoy receptor (IL-13Rα2). This receptor is overexpressed in many solid tumors but minimally expressed in normal tissues, making it an attractive target for tumor-specific immunotherapy. MDNA213 accumulates preferentially in the tumor microenvironment of IL-13Rα2-expressing cancers. It serves as a modular targeting domain for next-generation immunotherapies, including fusion proteins such as BiSKITs (Bifunctional SuperKines for ImmunoTherapy), where it can be fused to other therapeutic agents like anti-PD1 antibodies or cytotoxins to localize their activity within tumors. Preclinical studies have demonstrated that MDNA213-based constructs can enhance anti-tumor immune responses and exhibit potent, selective cytotoxicity against cancer cells expressing IL-13Rα2[2][6][7].
