MDNA213-PE is a novel Empowered Superkine in preclinical development by Medicenna Therapeutics. It is a recombinant fusion protein comprising MDNA213—an engineered Interleukin-13 (IL-13) Superkine with enhanced affinity for the Interleukin-13 receptor alpha 2 (IL-13Rα2)—fused to a truncated Pseudomonas exotoxin (PE). The drug is designed to selectively target and eliminate cancer cells that overexpress IL-13Rα2, a receptor associated with aggressive tumor phenotypes and poor prognosis in various cancers, including triple-negative breast cancer (TNBC). Upon binding to the receptor and subsequent internalization, the PE payload inhibits protein synthesis by ADP-ribosylating elongation factor 2, leading to cell death. Preclinical studies have demonstrated its potential as a monotherapy and its synergistic effects when combined with other immunotherapies like IL-2 Superkines.
