Overview

MDNA223 is a bifunctional fusion protein developed as part of Medicenna’s BiSKIT (Bifunctional SuperKines for ImmunoTherapy) platform. It consists of an engineered interleukin-2 (IL-2) "Superkine" fused to an anti-PD1 antibody. The drug is designed to activate cancer-killing immune cells by selectively stimulating the IL-2 receptor beta chain (IL-2Rβ), thereby promoting proliferation and activation of cytotoxic CD8+ T cells while minimizing stimulation of regulatory T cells. Simultaneously, it blocks PD-1 signaling on the same effector immune cell, preventing exhaustion and enhancing anti-tumor immunity through checkpoint inhibition. This cis-targeting approach allows both pathways to be modulated within a single molecule for improved efficacy in both immunologically “hot” and “cold” tumors. Preclinical studies have shown that MDNA223 demonstrates superior efficacy compared to separate administration of its components, with enhanced tumor inhibition and survival in multiple cancer models[1][2][4][5][6].
