Overview

**MDNA57** is a receptor-selective fusion protein engineered as an empowered cytokine by Medicenna Therapeutics, designed specifically to target the type II interleukin-4 receptor (IL-4R), which is overexpressed on solid tumor cells and supportive cells in the tumor microenvironment across over 20 cancer types. It combines an IL-4 superkine variant with a pro-apoptotic payload derived from Pseudomonas exotoxin to selectively induce apoptosis in type II IL-4R-expressing cells while sparing immune cells that predominantly express the type I IL-4R. As a second-generation iteration of Medicenna's IL-4 empowered cytokine platform (following MDNA55), MDNA57 aims to deliver potent anti-tumor cytotoxicity with improved selectivity for epithelial cancers and metastasis, addressing limitations of non-selective IL-4R targeting therapies.
