mDOF is a modified neoadjuvant chemotherapy regimen consisting of docetaxel, oxaliplatin, fluorouracil (5-FU), and leucovorin. It is specifically utilized in clinical trials conducted by the Sixth Affiliated Hospital, Sun Yat-sen University, for the treatment of advanced gastric cancer. The regimen is typically administered in cycles prior to D2 radical resection and may be combined with hyperthermic intraperitoneal perfusion chemotherapy (HIPEC). Docetaxel acts as a microtubule stabilizer, oxaliplatin functions as a platinum-based DNA cross-linking agent, fluorouracil is an antimetabolite that inhibits thymidylate synthase, and leucovorin is used to potentiate the effects of fluorouracil by stabilizing its binding to the target enzyme.
