mDR18 is an engineered murine decoy-resistant interleukin-18 (IL18) mutein delivered via a live bacterial platform. It is specifically designed to evade the inhibitory effects of the IL18 binding protein (IL18BP), which normally acts as a secreted immune checkpoint by neutralizing IL18. In this therapeutic system, mDR18 is expressed on the outer membrane of non-pathogenic *Escherichia coli* K-12 DH5α using bacterial scaffolds such as OmpA. This surface-display approach is intended to enhance the cytokine's effective concentration and half-life within the tumor microenvironment (TME) through surface anchoring and clustering. Preclinical studies demonstrate that mDR18-displaying bacteria induce robust CD8+ T cell and natural killer (NK) cell-dependent anti-tumor immune responses, promote immunologic memory, and exhibit abscopal effects in colorectal carcinoma and melanoma models.
