Drug intelligence / Profile preview
MDSY-1240 is an investigational, small-molecule proteolysis-targeting chimera (PROTAC) designed to selectively target and degrade signal transducer and activator of transcription 3 (STAT3). Developed by Medsyn Biopharma (a spinout from the University of Michigan), MDSY-1240 is being evaluated in a Phase 1/2 clinical trial for the treatment of relapsed or refractory hematologic malignancies and advanced solid tumors. These include triple-negative breast cancer, gastric cancer, ovarian cancer, head and neck squamous cell carcinoma, hepatocellular carcinoma, classical Hodgkin lymphoma, cutaneous T-cell lymphoma, peripheral T-cell lymphoma, and large granular lymphocytic leukemia. By recruiting an E3 ubiquitin ligase to STAT3, MDSY-1240 induces its polyubiquitination and subsequent degradation via the ubiquitin-proteasome system. This targeted degradation shuts down aberrant STAT3 signaling, which is a well-known driver of tumor cell proliferation, survival, metastasis, and immune evasion.
