MDT-637 (also known as VP-14637) is a small molecule antiviral drug developed as an inhibitor of the fusion protein of respiratory syncytial virus (RSV). It acts by blocking the RSV F protein, thereby preventing viral entry and fusion with host cells. MDT-637 demonstrated broad in vitro antiviral activity against multiple clinical and laboratory strains of RSV, with IC50 values ranging from 0.36 to 3.4 ng/mL[1][7][8]. The drug was formulated for inhaled delivery using a proprietary dry powder nebulizer to target the site of infection in the respiratory tract[6]. Clinical development included phase 1 and phase 2a trials assessing safety, tolerability, pharmacokinetics, and antiviral activity in healthy volunteers challenged with RSV[5][6]. Despite promising preclinical results showing superior potency compared to ribavirin, further development was discontinued for strategic reasons[2][4].
