Overview

KappaMab (MDX-1097) is a chimeric IgG1κ monoclonal antibody developed for the treatment of kappa light chain-restricted multiple myeloma and related B-cell malignancies. It specifically targets the kappa myeloma antigen (KMA), a membrane-bound form of free kappa light chain found on malignant plasma cells in kappa-type multiple myeloma and some lymphomas, but not on normal B cells or other healthy tissues. By binding to KMA, KappaMab induces immune-mediated killing of cancer cells through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). The drug has shown synergistic effects when combined with lenalidomide and dexamethasone in clinical trials for relapsed/refractory multiple myeloma. Developed by HaemaLogiX, it has demonstrated a favorable safety profile in Phase 1/2 studies and is being further evaluated in combination regimens.
