MDX-1203 is an investigational antibody-drug conjugate (ADC) developed for the treatment of cancers such as renal cell carcinoma and non-Hodgkin’s lymphoma. It consists of a fully human monoclonal antibody targeting the extracellular domain of CD70, a protein overexpressed in certain malignancies, chemically linked to a prodrug form of a CC-1065 (rachelmycin) analogue via a stable peptide-based linker. The cytotoxic payload is a DNA minor groove-binding agent that forms covalent adducts with adenine bases in DNA, thereby preventing DNA replication and inducing cell death. MDX-1203 was designed to selectively deliver its cytotoxic payload to tumor cells expressing CD70[1][5][6]. Development has been discontinued[2].
