MDX-124 is a novel, first-in-class humanized monoclonal antibody developed by Medannex that targets annexin-A1, a protein overexpressed in several cancer types. By binding to secreted and extracellular annexin-A1, MDX-124 disrupts its interactions with formyl peptide receptors (FPR1/2), preventing activation of downstream oncogenic processes that promote cancer cell proliferation and migration. It induces G1 phase cell cycle arrest, inhibits tumor growth in vivo, reduces metastasis, and triggers antibody-dependent cellular cytotoxicity (ADCC) in annexin-A1-expressing cancer cells. It is being developed primarily for the treatment of advanced solid tumors including pancreatic cancer (for which it has received US FDA Orphan Drug Designation), triple-negative breast cancer, colorectal cancer, and lung cancer.
