Overview

MDX-1342 is a fully human, nonfucosylated monoclonal antibody that targets the CD19 antigen, a membrane protein specifically expressed on normal and malignant B cells. By binding to CD19, MDX-1342 depletes and eliminates CD19-expressing B cells through enhanced antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis. The drug was developed for potential use in treating B-cell malignancies such as chronic lymphocytic leukemia (CLL), acute lymphoblastic leukemia, follicular non-Hodgkin lymphoma, diffuse large B-cell lymphoma, mantle cell lymphoma, as well as autoimmune diseases like rheumatoid arthritis. Preclinical studies demonstrated potent B-cell depletion and improved survival in animal models. Clinical development reached Phase 1 trials for CLL and rheumatoid arthritis but did not progress further[1][2][3][5][6][8].
