MDX-1401 is a fully human, second-generation, nonfucosylated monoclonal antibody that targets the cell surface receptor CD30. It was developed to treat CD30-positive malignancies such as Hodgkin lymphoma. The lack of fucose in its structure increases its affinity for Fc gamma receptor IIIa (CD16) on immune effector cells, resulting in enhanced antibody-dependent cellular cytotoxicity (ADCC) compared to earlier anti-CD30 antibodies. MDX-1401 has demonstrated preclinical and early clinical activity by reducing tumor burden and depleting circulating CD30-positive cells in patients with relapsed or refractory Hodgkin lymphoma. The drug was originally developed by Medarex and later by Bristol-Myers Squibb following their acquisition of Medarex[1][2][3][4][5][6].
