MDX2001 is a tetraspecific antibody-based T-cell engager (TeTE) developed by ModeX Therapeutics, an OPKO Health company. It is designed for the treatment of advanced solid tumors and various cancers including triple-negative breast cancer, prostate cancer, non-small cell lung cancer, pancreatic cancer, uterine cancer, colon cancer, gastric cancer, renal cell carcinoma, hepatocellular carcinoma, endometrial cancer, cervical cancer, esophageal cancer, biliary tract cancers and head and neck cancers. The drug targets four antigens simultaneously—CD3 (T cell surface glycoprotein), CD28 (T-cell-specific surface glycoprotein), c-MET (hepatocyte growth factor receptor), and TROP2 (tumor-associated calcium signal transducer 2)—to promote potent anti-tumor immune responses by engaging T cells with tumor cells expressing c-MET or TROP2. Preclinical studies show that MDX2001 induces strong cytolytic activity against a range of human tumor cell lines with minimal off-target activation of T cells in the absence of tumor antigens. The drug is administered intravenously[6][7][2][1].
