Overview

mDX400 is a murine-specific monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor. Developed by Merck & Co. (MSD), it serves as a preclinical surrogate for the human anti-PD-1 antibody pembrolizumab. It is widely used in syngeneic mouse tumor models to evaluate the efficacy of PD-1 blockade, either as a monotherapy or in combination with other agents such as histone deacetylase (HDAC) inhibitors (e.g., vorinostat) or BTLA antagonists. By blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), mDX400 reverses T-cell exhaustion and enhances anti-tumor immune responses in immunocompetent mice. Preclinical studies have demonstrated that mDX400 can significantly reduce tumor weight and lung metastases while prolonging survival in models of renal cell carcinoma, breast cancer, and colon cancer.
