ME-344 is a synthetic small molecule and an investigational antineoplastic agent that acts as a selective inhibitor of mitochondrial oxidative phosphorylation (OXPHOS), specifically targeting complex I. By inhibiting OXPHOS in tumor cells—especially when glycolysis is suppressed by antiangiogenic agents like bevacizumab—ME-344 disrupts ATP production and induces energy starvation. This leads to cancer cell death via both caspase-dependent and caspase-independent mechanisms including destructive autophagy. Preclinical studies have shown tumor-selective activity; clinical trials have evaluated its use in solid tumors such as ovarian cancer, small cell lung cancer, breast cancer (HER2-negative), and metastatic colorectal cancer. The drug was originally developed by Novogen and further developed by MEI Pharma[1][5][6][7].
