The measles DNA vaccine is an experimental DNA-based biologic designed to prevent measles infection, particularly in infants under six months of age who are ineligible for live attenuated vaccines due to maternal antibody interference. Developed by researchers at the Johns Hopkins School of Public Health and the Center for Vaccine Development in collaboration with Emory University, these vaccines utilize plasmid DNA to encode the measles virus hemagglutinin (H) and/or fusion (F) glycoproteins. Two primary candidates include pMSIN-H (encoding H alone) and pMSINH-FdU (encoding both H and F), often utilizing a Sindbis virus-based expression system. Some formulations incorporate the cationic lipid-based adjuvant Vaxfectin to enhance immunogenicity. The vaccine aims to induce both humoral and cellular immune responses against the measles virus surface proteins.
