mebendazole + temozolomide

Mebendazole + temozolomide is an investigational oral combination therapy consisting of mebendazole, a benzimidazole-class anthelmintic, and temozolomide, an alkylating agent. Mebendazole disrupts microtubule formation by binding to β-tubulin, leading to inhibition of cell division and induction of apoptosis in cancer cells. Temozolomide methylates DNA at the O6 and N7 positions of guanine residues, resulting in DNA damage and tumor cell death. This combination has been studied primarily for the treatment of high-grade gliomas (including glioblastoma), with clinical trials indicating acceptable safety profiles at high doses and potential improvement in survival outcomes compared to standard therapies alone[1][3][4][6].
