MED12 KO CAR NK cells are genetically engineered natural killer (NK) cell therapies in which the Mediator complex subunit 12 (MED12), a key component of the Mediator kinase module, is knocked out by CRISPR-based gene editing in NK cells that also express a chimeric antigen receptor (CAR) against tumor-associated antigens. Loss of MED12 enhances CAR‑NK cell antitumor activity by increasing metabolic fitness, expansion, and secretion of proinflammatory cytokines, and by promoting expansion of cytotoxic NK cell subsets, thereby augmenting both innate and CAR-mediated cytotoxicity against treatment-refractory cancers in vitro and in vivo.[3][6] These cells are being explored preclinically as next‑generation allogeneic cancer immunotherapies, including IL‑15–armored CAR‑NK constructs targeting antigens such as TROP2 and CD70.[3]
