Overview

MEDI-5304 is a fully human monoclonal antibody that functions as a dual neutralizing inhibitor of the Hedgehog signaling pathway by binding with high affinity to both sonic hedgehog (SHH) and Indian hedgehog (IHH) ligands. It blocks Hedgehog signaling in tumor-stromal interactions, demonstrating potent antitumor activity in preclinical models, particularly through the inhibition of paracrine Hedgehog signaling between tumor and stromal cells. MEDI-5304 has shown robust pharmacodynamic effects and antitumor efficacy in appropriate xenograft models but did not affect cancer stem cell frequency or growth in primary pancreatic cancer explant models. Adverse findings in preclinical toxicology were limited to odontodysplasia in rats. It is suitable for further development in human cancers with elevated SHH or IHH[1][3][4][5].
