Overview

MEDI-547 is an antibody-drug conjugate (ADC) developed for the treatment of solid tumors. It consists of a fully human immunoglobulin G1 monoclonal antibody (clone 1C1) targeting the cell-surface receptor Ephrin type-A receptor 2 (EphA2), which is overexpressed in various solid tumors and associated with poor prognosis. The antibody is conjugated to a cytotoxic auristatin derivative—maleimidocaproyl-monomethyl auristatin phenylalanine (mcMMAF)—which acts as a microtubule inhibitor by binding to the vinca alkaloid-binding domain. The unconjugated antibody has no antitumor effect; its purpose is to deliver the cytotoxic payload specifically to EphA2-expressing cancer cells, inducing apoptosis and inhibiting mitosis in these cells. Development was discontinued after early-phase clinical trials due to safety concerns related to bleeding and coagulation events[1][3][5][6].
