MEDI-570

MEDI-570 is a fully human, afucosylated IgG kappa monoclonal antibody that targets the inducible T-cell co-stimulator (ICOS), a protein highly expressed on activated CD4-positive T cells, including T follicular helper (Tfh) cells. By binding to ICOS, MEDI-570 blocks its interaction with the ICOS ligand (ICOSL) on plasmacytoid dendritic cells, leading to depletion of ICOS-expressing T cells and modulation of immune responses. The drug was developed by MedImmune (now part of AstraZeneca), originally from Japan Tobacco. It has been investigated primarily for relapsed/refractory angioimmunoblastic T-cell lymphoma (AITL) and peripheral T-cell lymphoma (PTCL), where it showed promising clinical activity and tolerability in early-phase trials. Development for systemic lupus erythematosus was discontinued after phase 1[1][2][3][4][5][6].
