MEDI0457 is a DNA-based cancer vaccine and immunotherapy designed to treat cancers associated with human papillomavirus (HPV) types 16 and 18[9][4][14][11]. It consists of two main plasmid components: VGX-3100, which encodes modified HPV E6 and E7 oncoproteins, and INO-9012, which encodes interleukin-12 (IL-12) as an immune adjuvant[4][9][14]. Upon intramuscular injection and electroporation, these plasmids trigger cellular expression of E6 and E7 proteins and IL-12, leading to strong cytotoxic T-lymphocyte and NK cell activation, thereby targeting and lysing HPV-infected tumor cells. In clinical trials, MEDI0457 has shown enhanced cellular and humoral immune responses and is evaluated in combination with durvalumab (an anti-PD-L1 monoclonal antibody) for recurrent/metastatic HPV-positive cancers, including head and neck, cervical, anal, penile, and vulvar cancers[1][3][5][14].
