**MEDI0562 + durvalumab** is a combination immunotherapy regimen under investigation for the treatment of advanced solid tumors. MEDI0562 is a humanized IgG1κ monoclonal antibody that acts as an agonist of OX40, a costimulatory molecule expressed on activated T cells, enhancing antitumor immune responses. Durvalumab is a human monoclonal antibody targeting programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 to restore T-cell function. This combination aims to both stimulate effector T cell activity (via OX40 agonism) and relieve immune suppression (via PD-L1 blockade) to achieve enhanced antitumor effects. Early-phase clinical studies have shown immunomodulatory activity but have not established clear efficacy signals. The combination is being developed primarily for previously treated advanced solid tumors, using intravenous administration. The primary developer is MedImmune/AstraZeneca[1][3][7][9].
