Overview

MEDI0680 is a humanized immunoglobulin gamma 4 kappa (IgG4κ) monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor on T cells. By binding to PD-1, it blocks the interaction with its ligands PD-L1 and PD-L2, which are often exploited by tumor cells to evade immune detection. This blockade restores and enhances T-cell activation and antitumor immune responses, making it an immune checkpoint inhibitor with potential anticancer activity. MEDI0680 was developed for the treatment of various cancers, including advanced solid tumors such as renal cell carcinoma and melanoma[3][4][5]. It has been investigated both as monotherapy and in combination with other checkpoint inhibitors like durvalumab[5][7].
