**MEDI0680 + durvalumab** is a combination of two monoclonal antibodies: MEDI0680, a humanized anti–programmed cell death-1 (PD-1) antibody, and durvalumab, an anti–programmed death-ligand 1 (PD-L1) antibody. This combination is intended to enhance antitumor immune responses by dual inhibition of the PD-1/PD-L1 pathway, which promotes immune activation against tumors. MEDI0680 blocks the interaction between PD-1 and its ligands, thus restoring and improving intrinsic T-cell function, while durvalumab inhibits PD-L1-mediated immune evasion by tumor cells. The combination has been primarily studied in patients with advanced clear-cell renal cell carcinoma and other advanced solid malignancies.
