Overview

**MEDI1191 + durvalumab** is a combination investigational therapy consisting of **MEDI1191**, a lipid nanoparticle-formulated messenger RNA (mRNA) encoding human interleukin-12 (IL-12), and **durvalumab**, an anti-PD-L1 monoclonal antibody. MEDI1191 is administered via intratumoral injection and promotes local production of IL-12 in the tumor microenvironment, thereby driving antitumor immunity through the activation of multiple immune effector mechanisms, such as inducing IFNγ release and increasing CD8+ T cell tumor infiltration. Durvalumab, given intravenously, blocks PD-L1, preventing immune evasion by tumor cells and enhancing immune responses. The combination is under investigation for advanced solid tumors in phase 1 clinical trials, showing preliminary evidence of safety, immune activation, and antitumor activity in patients with multiple cancer types, including melanoma, breast cancer, colorectal cancer, head and neck cancer, and more. Development and studies are sponsored by MedImmune, a subsidiary of AstraZeneca[1][2][5][6].
