Overview

MEDI1341 is a human IgG1λ monoclonal antibody engineered for high-affinity and high-selectivity binding to both monomeric and aggregated forms of alpha-synuclein. It features reduced effector function to minimize immune system interactions. The drug is designed to sequester extracellular alpha-synuclein and block its cell-to-cell transmission in the brain, aiming to slow or halt the progression of synucleinopathies such as Parkinson’s disease and multiple system atrophy. Preclinical studies demonstrated that MEDI1341 enters the central nervous system after intravenous administration, lowers free alpha-synuclein levels in cerebrospinal fluid and brain interstitial fluid, and reduces propagation of pathological alpha-synuclein in animal models. Clinical development is ongoing with Phase 2 trials for multiple system atrophy and Phase 1 trials for Parkinson’s disease[4][5][6][7][8].
