MEDI1341-D265A is an effector-null variant of the monoclonal antibody MEDI1341, developed by AstraZeneca in collaboration with Takeda Pharmaceutical Company. It is a fully human IgG1 monoclonal antibody that binds to the C-terminal region of Alpha-synuclein, targeting both monomeric and aggregated forms of the protein. The D265A mutation in the Fc region specifically eliminates binding to Fcγ receptors, thereby removing effector functions such as microglial uptake while retaining the antibody's ability to sequester extracellular Alpha-synuclein. This mechanism is intended to prevent the cell-to-cell transmission and seeding of pathological Alpha-synuclein, a process believed to drive the progression of alpha-synucleinopathies. MEDI1341-D265A is being developed for the treatment of Parkinson's disease and multiple system atrophy, with the aim of slowing or halting disease progression.
