Overview

MEDI1814 is a fully human high-affinity monoclonal antibody that selectively targets the C-terminus of amyloid-beta 42 (Aβ42), a pathogenic self-aggregating species implicated in Alzheimer's disease. Developed originally by MedImmune (now part of AstraZeneca) and co-developed with Eli Lilly, this IgG1λ antibody is designed to reduce free Aβ42 in the cerebrospinal fluid without affecting Aβ40 levels. Its mechanism involves binding monomeric Aβ42 and inhibiting its aggregation, aiming to slow or modify disease progression in Alzheimer's disease. The drug has shown favorable safety and tolerability profiles in early clinical studies and is being investigated as a potential disease-modifying therapy for mild to moderate Alzheimer's disease[1][2][3][6][7][8].
