Overview

MEDI3039 is a novel, highly potent, multivalent tumor necrosis factor (TNF)-related apoptosis-inducing ligand receptor 2 (TRAIL-R2 / DR5) agonist. It is an engineered multivalent scaffold protein based on the third fibronectin type III domain of the human glycoprotein tenascin C, linked to polyethylene glycol-20 (PEG20) to improve in vivo half-life. MEDI3039 is designed to induce apoptotic cell death in cancer cells by activating death receptor 5 (DR5) with high potency and without requiring Fcγ receptor-mediated cross-linking. It has demonstrated significant preclinical efficacy in solid tumors, particularly triple-negative breast cancer (TNBC) and colorectal cancer.
