MEDI3185 is a fully human monoclonal antibody that acts as a potent antagonist of the C-X-C chemokine receptor type 4 (CXCR4). Developed by MedImmune (a subsidiary of AstraZeneca), it blocks the binding of the ligand stromal-derived factor 1 (SDF-1/CXCL12) to CXCR4, thereby inhibiting downstream signaling pathways involved in tumor cell migration, survival, and proliferation. In preclinical studies, MEDI3185 demonstrated anti-tumor activity as a single agent in models of multiple myeloma and Burkitt's lymphoma, and showed synergistic effects when combined with standard therapies in ovarian cancer and chronic lymphocytic leukemia (CLL) models. The antibody exhibits picomolar binding affinity to human CXCR4 and does not show significant binding to other chemokine receptors such as CCR4 or CXCR3.
