Overview

MEDI3617 is a fully human IgG1 kappa monoclonal antibody that selectively binds to angiopoietin 2 (Ang2) with high affinity, inhibiting its interaction with the TEK tyrosine kinase receptor (Tie2). This blockade disrupts Ang2-induced Tie2 phosphorylation, thereby inhibiting angiogenesis and tumor vascular remodeling. MEDI3617 was developed as an antiangiogenic agent for the treatment of advanced solid tumors, including ovarian cancer and glioma. Preclinical studies demonstrated its ability to inhibit tumor growth in various xenograft models. Clinical trials evaluated it both as monotherapy and in combination with bevacizumab or chemotherapy in patients with advanced solid tumors refractory to standard therapy[1][2][5]. Development was discontinued due to limited clinical activity and safety concerns, particularly prolonged grade 3 edema-associated adverse events observed in ovarian cancer patients[5].
