Overview

MEDI3622 is a human monoclonal antibody developed by MedImmune (a subsidiary of AstraZeneca) that targets ADAM17 (a disintegrin and metalloprotease 17), also known as TNF-alpha converting enzyme (TACE). ADAM17 is a membrane-bound protease responsible for the proteolytic cleavage or 'shedding' of various cell-surface proteins, including TNF-alpha and ligands for the epidermal growth factor receptor (EGFR) such as TGF-alpha and amphiregulin. By inhibiting ADAM17, MEDI3622 blocks the release of these pro-tumorigenic and pro-inflammatory factors, thereby disrupting autocrine and paracrine signaling pathways like EGFR and STAT3. Preclinical studies in pancreatic ductal adenocarcinoma (PDAC) models have shown that MEDI3622 can induce tumor regression, increase cytotoxic T-cell infiltration, and reduce the presence of granulocytic myeloid-derived suppressor cells (G-MDSCs). Although it entered Phase 1 clinical trials for advanced solid tumors, the clinical development program was subsequently terminated.
