Overview

MEDI4166 is a recombinant fusion protein consisting of a glucagon-like peptide-1 (GLP-1) analogue fused to human alpha-1 antitrypsin (AAT). Developed by MedImmune, a subsidiary of AstraZeneca, the molecule was designed as a potential treatment for type 2 diabetes mellitus. The GLP-1 component acts as an agonist at the GLP-1 receptor, promoting glucose-dependent insulin secretion and suppressing glucagon release. The AAT component, a serine protease inhibitor, provides anti-inflammatory and cytoprotective properties intended to protect pancreatic beta cells from inflammatory stress and apoptosis. This dual-action approach aimed to achieve both glycemic control and disease modification by preserving beta-cell function and mass. Clinical development reached Phase 1, but the program was subsequently discontinued.
