Overview

MEDI5083 is a novel, engineered fusion protein designed as an agonist of the CD40 pathway for cancer immunotherapy. It consists of a hexameric recombinant human CD40 ligand (CD40L) structure covalently linked to a human IgG4P Fc domain, forming a trimerized IgG fusion protein that mimics the native ligand for enhanced potency. MEDI5083 specifically binds and activates CD40, a receptor on antigen-presenting cells such as B lymphocytes, monocytes, and dendritic cells. Activation of this pathway bridges innate and adaptive immunity through direct tumor cell apoptosis, cytotoxic T-cell mediated killing, and activation of the tumor microenvironment via M1 polarization. Developed for subcutaneous administration to prolong exposure and minimize systemic toxicity, MEDI5083 was evaluated in phase 1 clinical trials in combination with durvalumab across multiple solid tumors but further development was limited by its toxicity profile[1][2][5][6].
