MEDI5083 + durvalumab is an experimental combination therapy evaluated for advanced solid tumors. MEDI5083 is a CD40 agonist fusion protein designed to stimulate the CD40 receptor on antigen-presenting cells, enhancing immune activation against tumor cells. Durvalumab is a selective, high-affinity, engineered human IgG1 monoclonal antibody that blocks PD-L1 from binding to PD-1 and CD80, thereby facilitating T-cell activation and tumor cell elimination. The therapeutic rationale of this combination is to synergistically activate both innate and adaptive immune responses, potentially overcoming PD-L1-mediated resistance seen with CD40 activation alone. Clinical testing was discontinued due to toxicity and inability to deliver biologically efficacious doses.
