MEDI5884 is a humanized immunoglobulin G4P kappa monoclonal antibody that selectively neutralizes endothelial lipase (EL). By inhibiting EL, which hydrolyzes phospholipids and promotes the catabolism of high-density lipoprotein (HDL), MEDI5884 increases both the quantity and quality of HDL particles. This mechanism facilitates reverse cholesterol transport, potentially offering cardioprotective effects by enhancing cholesterol efflux from tissues to the liver for excretion. In clinical studies, MEDI5884 has demonstrated dose-dependent increases in HDL cholesterol and global cholesterol efflux in patients with stable coronary artery disease on statin therapy. The drug was originally developed by AstraZeneca and later licensed to Regio Biosciences for further development in cardiovascular indications such as peripheral artery disease[1][6][8].
