Overview

MEDI7219 is a synthetic peptide drug developed as a glucagon-like peptide 1 receptor (GLP-1R) agonist for the treatment of type 2 diabetes. It was designed to be orally administered and features side-chain lipidation to increase its half-life via reversible albumin binding, as well as the incorporation of unnatural amino acids to enhance proteolytic stability. The drug acts by stimulating insulin secretion in response to food intake, thereby lowering blood glucose levels. Preclinical studies demonstrated similar potency to semaglutide *in vitro* and significant glucose-lowering effects in animal models. Clinical development reached phase 1 trials but was discontinued before further advancement.
