MEDI7247 is a first-in-class antibody-drug conjugate (ADC) designed for the treatment of hematological malignancies and selected solid tumors. It consists of a human monoclonal antibody targeting the sodium-dependent alanine-serine-cysteine transporter 2 (ASCT2, also known as SLC1A5), which is overexpressed in various cancers. The antibody is site-specifically conjugated via a protease-cleavable linker to a highly cytotoxic pyrrolobenzodiazepine (PBD) dimer payload, which acts as a DNA cross-linking agent. This design enables targeted delivery of the cytotoxic agent to ASCT2-expressing tumor cells while sparing normal tissues with low ASCT2 expression. MEDI7247 has been evaluated in phase 1 clinical trials for relapsed or refractory acute myeloid leukemia (AML), multiple myeloma (MM), diffuse large B-cell lymphoma (DLBCL), and advanced/metastatic solid tumors[1][5][6].
