MEDI7814

MEDI7814 is a human immunoglobulin G4 (IgG4), effector-null, neutralizing monoclonal antibody that specifically targets human complement component C5a and its des-Arg derivative (C5adesArg). It inhibits the binding of C5a and C5adesArg to both the Complement component 5a receptor 1 (C5aR1) and Complement component 5a receptor 2 (C5aR2) receptors, thereby blocking downstream inflammatory signaling. Unlike other anti-C5a monoclonal antibodies, MEDI7814 blocks both receptor pathways without affecting complement-mediated bacterial cell killing. The drug was developed for acute inflammatory conditions where both receptors mediate inflammation, such as sepsis or renal ischemia–reperfusion injury. It was also investigated for chronic obstructive pulmonary disease but development in this indication has been discontinued[1][2][3][4][5][6].
