MEDI8111 is a recombinant human prothrombin (rhFII), developed as a thrombin modulator to activate the blood coagulation pathway. It is designed to increase procoagulant activity and reduce bleeding in conditions such as hemophilia A and B. Preclinical studies have shown that MEDI8111 dose-dependently decreases bleeding and increases thrombin generation in animal models of hemophilia. The drug acts by supplementing or replacing deficient or inhibited endogenous prothrombin (Factor II), thereby enhancing thrombin generation and promoting hemostasis[2][4]. Developed initially by MedImmune (a subsidiary of AstraZeneca), its primary indication is for the prevention of bleeding episodes in patients with hemophilia A and B who may have developed inhibitors to standard replacement therapies[2][4].
