Overview

MEDI8852 is a human IgG1 kappa monoclonal antibody that targets a highly conserved region within the stalk of the hemagglutinin (HA) protein of influenza A viruses. By binding to this region, it neutralizes and prevents essential steps in the viral lifecycle, including fusion between HA and cellular membranes, HA protein maturation, and host cell protease cleavage. This action blocks infectivity across all influenza A virus subtypes. Additionally, MEDI8852 facilitates clearance of infected cells through Fc effector functions such as antibody-dependent cellular cytotoxicity, phagocytosis, and complement-dependent cytotoxicity. It has demonstrated broad neutralizing activity against seasonal H1N1 and H3N2 viruses as well as other subtypes with pandemic potential (H2, H5, H6, H7, H9). The drug was initially developed by Humabs (Switzerland) using human memory B cells from recovered patients and later further developed by MedImmune/AstraZeneca for treatment of influenza A infection[1][2][3][4][8].
