Overview

MEDI9253 is a novel, modified oncolytic virus based on recombinant Newcastle disease virus (NDV) that encodes the human interleukin-12 (IL-12) gene. It is designed to selectively replicate in tumor cells, leading to direct tumor cell lysis and local expression of IL-12 within the tumor microenvironment. The expressed IL-12 acts as an immune stimulatory cytokine, activating both innate and adaptive immune responses against cancer cells. MEDI9253 was developed for use in advanced or metastatic solid tumors and has been studied in combination with durvalumab (a PD-L1 checkpoint inhibitor). The drug was initially developed by AstraZeneca and reached phase 1 clinical trials before being discontinued for solid tumors due to limited efficacy[1][4][7].
