Adjuva Bio is developing a fibroblast-directed cell-engineering platform that utilizes lipid nanoparticle (LNP) encapsulated mRNA to simultaneously overexpress Meflin (ISLR) and a decoy IL-33 receptor (dST2). Meflin is a protein that marks anti-fibrotic fibroblast subsets and inhibits myofibroblast differentiation and collagen deposition. The decoy ST2 receptor (dST2) acts by sequestering IL-33, an alarmin that promotes type-2 (Th2) inflammation and pro-fibrotic fibroblast activation. By combining these two mechanisms, the therapy aims to reprogram pathogenic fibroblasts and restore tissue homeostasis in idiopathic pulmonary fibrosis (IPF) and other fibro-inflammatory diseases.
