Overview

Megakaryocyte-derived extracellular vesicles (MkEVs) are a specialized class of biologic therapeutic agents and cell-derived nanoparticles. These vesicles are typically derived from megakaryocytes, which can be generated through the differentiation of human pluripotent stem cells (hPSCs). MkEVs possess a natural tropism for hematopoietic stem and progenitor cells (HSPCs) within the bone marrow, a property that enables them to serve as highly efficient delivery vehicles for therapeutic cargoes, including gene editing tools, mRNA, and small molecules. Beyond their utility in drug delivery, MkEVs have been shown to directly stimulate thrombopoiesis, leading to increased platelet counts. They are currently being investigated for the treatment of a wide range of conditions, including genetic disorders, infectious diseases, and hematopoietic disorders such as thrombocytopenia, anemia, and hemoglobinopathies.
