Melan-a analog peptide is a synthetic modified decapeptide derived from the melanocyte differentiation antigen Melan-A/MART-1. The most studied variant is the A27L substitution (ELAGIGILTV), which replaces alanine with leucine at position 27 of the native sequence. This modification increases its binding affinity to HLA-A*0201 and enhances immunogenicity compared to the native peptide (EAAGIGILTV). The peptide acts as an antigenic epitope recognized by CD8+ cytotoxic T lymphocytes in an HLA-A2-restricted manner. It has been used in cancer immunotherapy research—particularly for melanoma and multiple myeloma—by stimulating T-cell responses against tumor cells expressing Melan-A/MART-1 or cross-reactive antigens such as HM1.24. Clinical studies have shown that vaccination with this analog induces stronger and more frequent T cell responses than the natural peptide, making it a promising candidate for cancer vaccine development[1][2][6][7][9].
