Melan-A ELA + Montanide is a therapeutic cancer vaccine formulation investigated for the treatment of malignant melanoma. It consists of a synthetic analog of the Melan-A/MART-1 tumor-associated antigen, specifically the ELA peptide (sequence: ELAGIGILTV), which is modified from the natural EAA sequence to enhance its binding affinity to the HLA-A*0201 molecule. This peptide is emulsified with a Montanide adjuvant (typically Montanide ISA 51 or ISA 720), a water-in-oil emulsion that creates a depot effect at the injection site. This formulation facilitates the slow release of the antigen and promotes a robust cytotoxic T-lymphocyte (CTL) response against melanoma cells expressing the Melan-A antigen. Developed primarily through academic and research institutions like Lausanne University Hospital and the Ludwig Institute for Cancer Research, the vaccine has been evaluated in Phase I and Phase II clinical trials, often in combination with other tumor antigens and immunomodulatory agents such as CpG or IL-2.
